This web page contains data files supplemental to:
"Characterization of synthetic DNA bar codes in Saccharomyces cerevisiae gene-deletion strains". Robert G. Eason, Nader Pourmand, Waraporn Tongprasit, Zelek S. Herman, Kevin Anthony, Olufisayo Jejelowo, Ronald W. Davis, and Viktor Stolc. Proc Natl Acad Sci U S A. 2004 Jul 27;101(30):11046-51

There are 5 tables containing the analysis result:

Table1 (Table1.txt, 605 KB)

This table contains the designed barcode sequences and the barcodes sequences found in the deletion cassettes for uptag and downtag of 5934 ORFs. The designed barcodes sequences are all denoted in capital letters. If the barcode sequence found in the deletion cassette has the same identity as the designed barcode, then the sequence is denoted in capital letters. The barcode sequence found in the cassette with error(s) is denoted in lower case letters. The causes of errors can be:

This table is provided in tab-delimited format.

Table2 (Table2.xls, 2.3 MB)

This table contains the summary of all types of mutation in the common primer and the unique barcode sequences for uptags and downtags of 5934 ORFs. Deletion is denoted with 'D', insertion is denoted with 'I', and substitution is denoted with 'M'.

This table is provided in MS Excel format.

Table3 (Table3.xls, 657 KB)

This table contains 760 ORFs that are considered to have anomalous UPTAG hybridization signals and 845 ORFs that are considered to have anomalous DNTAG hybridization. The LR values for the 760 ORFs are less than “normal” hybridization (i.e., below +/-3 SD of the average), and the LR values for the 845 ORFs are greater than “normal” hybridization (i.e., above +/-3 SD of the average).

This table is provided in MS Excel format.

Table4 (Table4.xls, 36 KB)

This table contains the percentage of ORF population with defect(s) that have LR outside the normal range.

Table5 (Table5.xls, 92 KB)

This table contains 182 ORFs that would be excluded from the analysis because the hybridization intensities of both tags are below background (or one tag is below background if the deletion strain has only one barcode).
